THE SCIENCE
PT-141, also known as bremelanotide, is a synthetic peptide analogue of alpha-melanocyte-stimulating hormone (α-MSH) that acts on melanocortin receptors. Research has focused primarily on its effects on sexual desire, sexual arousal, and erectile response through central melanocortin signaling.
 melanocortin signaling.
PT-141 acts as an agonist at melanocortin receptors, including MC3R and MC4R. These receptors are expressed in the central nervous system and are involved in signaling pathways associated with sexual motivation, arousal, and erectile responses. PubMed
SEXUAL DESIRE & AROUSAL
Human research has investigated PT-141 in relation to sexual desire and arousal, particularly in premenopausal women with reduced sexual desire or sexual arousal. Clinical studies have reported improvements in measures of sexual desire compared with placebo under the conditions studied.
ERECTILE RESPONSE
PT-141 has also been studied in men in relation to erectile response. Clinical research in healthy men and men with erectile dysfunction has reported increased erectile activity following PT-141 administration compared with placebo
CENTRAL SEXUAL SIGNALING
Research suggests that melanocortin signaling can influence neural pathways involved in sexual motivation and arousal. Experimental studies of PT-141 have examined activity in hypothalamic and other central nervous system regions associated with sexual behavior and response. PubMed
HUMAN CLINICAL RESEARCH
- Bremelanotide has been evaluated in randomized clinical trials involving sexual dysfunction. Phase 3 studies in premenopausal women with hypoactive sexual desire disorder reported statistically significant improvements in sexual desire and reductions in distress related to low desire compared with placebo. Earlier clinical research also examined erectile responses in men
RESEARCH LIMITATIONS
Research findings for PT-141 should be interpreted according to the study population, formulation, route of administration, experimental design, and specific endpoints measured. Clinical evidence has focused on particular sexual-function populations and does not establish that every PT-141 preparation has the same characteristics or outcomes.
